Pharmacokinetics
Tinidazole has a longer half life than Metronidazole and is better tolerated. Its t1/2 is greater than 12 hrs, hence used for single dose therapy. It is metabolised in the liver through glucuronidation and Oxidation and is excreted in the urine.
Indications
Tinidazole is given for treatment of protozoal infections like amoebiasis, giardiasis, trichomoniasis . It is also used in the treatment of anaerobic infections.
Routes of Administration and Dosage
For oral dosage form: For Amoebiasis in adults: 2gms once daily for 3 days, in children 30-50mgs/kg/day for 3 days. For giardiasis and trichomoniasis: 2gms single dose or 0.6gm once daily for 7 days. For anaerobic infection : Prophylactic in adults: 2gms
Contra Indications
Tinidazole is contraindicated in blood dyscrasias, active neurological diseases and in first trimester of pregnancy.
Precautions
Precaution has to be taken in late trimesters of pregnancy, seizure disorders, in hepatic and renal impairment and in alcoholics.
Interactions
Tinidazole increases the risk of bleeding when combined with anticoagulants. When the drug is taken along with alcohol, disufiram like reaction occurs. Enzyme inducers ( phenobarbitone, rifampicin ) and cimetidine can reduce the metabolism of tinidazole.
