Pharmacokinetics
Griseofulvin is given orally in divided doses to attain maximum level in the blood. It is mainly excreted through faeces.Because of its very low water solublity absorption from G.I.T is irregular.Absorption is increased by taking it with fat and by microfining the drug particles.Griseofulvin gets deposited in keratin forming cells of skin, hair and nails.It is specially concentrated and retained in tinea infected cells.
Indications
Griseofulvin is used to treat tinea infections of the skin, hair, fingernails, and toenails. It is used systemically only against Trichophyton, Epidermophyton and Microsporum.It is ineffective topically.
Routes of Administration and Dosage
Oral;125 to 250 mgs q.i.d with meals.Duration of treatment depends on the site of infection.Body skin - 3 weeks,palms and soles 4 to 6 weeks,finger nails 4 to 6 months, toe nails 8 to 12 months.
Contra Indications
Griseofulvin is contraindicated in severe hepatic disease, pregnancy, porphyria, monilial infection and SLE.
Precautions
Griseofulvin may increase the effects of alcohol. If taken with alcohol it may also cause tachycardia, increased sweating, flushing of the face.
Interactions
Griseofulvin may decrease the effect of anticoagulants in some patients. While taking Oral contraceptives containing estrogen with Griseofulvin, the effect of contraception is decreased, which may result in breakthrough bleeding and unwanted pregnancies.
