Pharmacokinetics Ceftriazone is a long acting third generation cephalosporin, with half life of 8 hours. It has a good CSF penetration and eliminated equally in urine and bile. Indications Ceftriazone is used in bacterial meningitis[espcially in children], multi-resistant typhoid fever, complicated UTI, abdominal sepsis and septicemia. A single dose of 250 mgs I.M has proved curative in gonorrhea including PPNG. Routes of Administration and Dosage IN meningitis; 4gms followed by 2gms I.V[children 75-100mgs\kg] once daily for 7-10 days. Typhoid; 4gms I.V for 2 days followed by 2 gms\day[for children 75 mgs\kg] till 2 days after the fever subsides. Contra Indications Ceftriazone is contraindicated in patients with hypersensitivity to cephalosporins. Precautions Hypoprothrombinemia and bleeding are specific adverse effects of cephalosporins. Precautions are to be taken in renal failure. Interactions Ceftriazone gives Disulfiram like reaction with alcohol.
