Pharmacokinetics
It is a proton pump inhibitor.It is rapidly absorbed.It supresses the gastric acid secretion by inhibition of Hydrogen Potassium ATPase and thus blocks the final step in gastric acid secretion. It causes almost 100% inhibition of gastric acid secretion in the stomach. It has a plasma 1/2 life of 30-90 min, and duration of action is 24 hours. It is 98 % bound to plasma proteins. It is metabolised in liver and excreted mainly in the urine.
Indications
It is used in the treatment of Duodenal ulcer, Gastric ulcer, Zollinger Ellison syndrome and Reflux oesophagitis.
Routes of Administration and Dosage
For peptic ulcer: 20-40 mg/day four to eight weeks; Zollinger Ellison syndrome: 60 mg /day(20 to 120 mgs/day); reflux oesophagitis: 20-40 mg /day for four to eight days.
Contra Indications
It is contraindicated in persons with hypersensitivity to the drug,neonates and during lactation.
Precautions
Precautions must be taken when taking the drug during pregnancy,cirrhosis, malignancy and prolonged use.
Interactions
It prolongs the elimination of diazepam, warfarin, phenytoin and aminophylline.
