Pharmacokinetics
Nalidixic Acid when given orally is readily absorbed from the GIT, 80% is eliminated through the kidneys. Both active & inactive forms of Nalidixic acid is found in the urine.Concentration in urine is 20 to 50 times that in plasma.
Indications
Nalidixic acid is used to treat infections of the urinary tract. It is highly effective against Gram negative Bacilli like E.Coli, Proteus. It has a weak action against Klebsiella and Aerobacter.Has a special place in treating ampicillin resistant shigella enteritis.
Routes of Administration and Dosage
500 mgs to 1 gram every six to eight hours.
Contra Indications
It is mainly contraindicated in infants upto 3 months of age and in those with convulsive disorders.
Precautions
Adverse effects : the most important toxicity is neurological toxicity such as headache, drowsiness, vertigo, visual disturbances and occasionally seizures.Prolonged use has produced parkinsonism like symptoms, leukopenia and biliary stasis. When used in individuals with G6PD deficiency, they may develop hemolysis.
Interactions
Interaction occurs with Anticoagulants : Patients taking Nalidixic acid with anticoagulants may have an increased chance of haemorrhage.Nitrofurantoin should not be used concurrently.
